Ads on Twitter.com would be fine, ads in the stream are not. I don't want to pay to receive ads, that's crazy.
Why not let businesses buy keywords and then have those keywords link back to a site? For example, I could buy "fitness video" and whenever someone typed in "fitness video" that text would link to http://www.beYOU.tv (where I work).
This is a non-intrusive and original way to generate money for Twitter.
What do you think?

Do I prefer and ad-free Twitter? You bet. Would I stop using if there were ads? Probably not.
Ads do seem the easy way out. And I love people who impress me with their creativity...

They're seeing a ton of interesting data flow through their servers, they could start making sense of it and selling it. Of course the privacy aspects are a bit scary, but if they do a good job of handling that I think there's a lot of potential upside.
SMSGupshup (www.smsgupshup.com) has been providing a similar service in INdia. Do try it out if you are in India.
Needless to say, I work at SMSGupshup - would love to know your comments, feedback, brickbats. pratyush [at] webaroo[dot] com
